After you applied this product to a pipe thread and placed the nut, do you have to wait, or can you start running the water right away?

Depends on how much pressure there is. I know if you put it on a airhose fitting and then pressurize it right away, it won't hold and start to push it out of the threads. I think it takes 24hrs to setup fully.

Use on drinking water systems?

Not sure, I used it on air lines outside. I would use Teflon tape on water.

Can one use teflon tape, then apply this product on top of the tape (for water pipe threads)?

You would use one or the other, you wouldn't use both... this would take the place of teflon tape.

Can this be used on a high pressured air mattress hose? I need something to seal the threads, but not close off the intake/outtake capacity of hose.

Yes, you should be able to use it on air mattress hose, as long as they're threaded and not just push-on type connections
