Does it fit a janome skyline s5

Yes it does. It is the machine I bought my Tutto for. It's a very tight fit, but it works, even though there is no room to spare.

will this bag accommodate a Singer 8500Q modern quilter?

That looks like the same machine my fiance has. It fits nicely inside the bag.

I haven't seen this answered anywhere. When machine is loaded are there side handles for removing/lifting from car?

no, you only have the handle on top of the bag. I haven't found this to be a problem with taking the machine out of my Ford Explorer back seat. I love the bag. There's plenty of storage pockets and moves around so easily. I also love the fact you can collapse it down when not in use for easy storage. Hope this helps.
