How many uses can you get out of the tape? Do the tiles seem to stop sticking after just a few play throughs?

They do stop sticking after a few play throughs. The tape is easily replaced. But, I've also used them without the tape. My group tends to plow through areas, so for us it is often easier to use without the tape so it's more easily adjusted.

Would I be able to attach magnets to the bottom or even some crews or nuts to add weight to not need the tacky sticker to keep them upright?

Screws no, magnetic tape yes. You would need a ferrous metal sheet under your map though. The blocks are made of high density foam.

Is it possible to purchase more of the suction tape that comes with the set?

Definitely, probably not from the same company but you can definitely find double sided tape on Amazon
