Are these stickers easy to remove after they are applied to an item?

Depends on how long stuck there and surface. If haven’t been very long, come off fairly easily on any surface. If have been there for a while, they don’t come off as well, not wood hardly at all and plastic about half the time.

I'm wondering if these would work for line spots in my classroom if I adhere them to the ground?

No, they are way too small for that. They are about the size of like those garage sale stickers if you know what I’m talking about. Or like a normal smiley face sticker that you’d put on a students paper.
